So it's a bit like responsive web design (it loads lighter images/content to speed up load time and reduce CPU %)? I'll try adopting freezing & let you know how it goes. Personally, I don't do it this way as I tend to work within the limits of my machine and do all the mixing within the project, just at the very end of production. It also shifts focus from wanting to make further edits but if you do need to, you can just export the stems again and re-import into your "mixing" project and have all the same processing applied (being cautious about any automation) What you then do is either reload those as a new project in Live if you like to stay in Live but many will take these to a more "traditional" linear DAW where they can then focus purely on the mixing side where you now have all the power available for processing the sound to the final mix. That will generate a wav file for each separate track often referred to as "stems". You can choose in the Live Export options to render instead of the Master, "All Tracks": Should sound identical to how it was tracked. Freeze will just create an in place render and place all the devices "offline" so a "representation" of that track is played instead. Thanks!įreeze can be useful when you are producing and a heavy track is mostly done but takes a lot of processing. Once you get pass lets say 6db of compression is when it doesnt sound as good. ![]() I love it for 1 to 3db of compression as its very transparent. When you put L1 on, it changes the sound as well as 元. These options seem like good suggestions. Well, believe it or nah, L2 is the cleanest of them all. Tue 10:37 - Is there a risk with rendering tracks where you can't go back and easily make changes? Or when it is rendered, is it dynamic? Same question for freezing. It's about working smarter, not just increasing the CPU and demanding more efficiency for the way you choose to work, look into how seasoned professionals work, i'm not one of them but I take notes from their books. ![]() Render tracks out and mix/master stems instead of doing all within your production environment.Ī reason some will mix in a different program, apart from a more desirable workflow, you can drop all the load from synths and track plugins and switch to mixing plugins and devote all the processing for the master section. Load is how many CPU cycles all your software and plugins will allow for, latency is the delay in time needed for plugins to calculate their process. To achieve this, use Pro-L 2 on the stem channel, while feeding the original (unmastered, unlimited) master to its side chain input. Any ideas?ĭepends on system and specs (not stated). This can be very useful for stem mastering, when you need to deliver individual stems that have been processed with the exact same limiting as the original master. There has to be a way around this for producers. ![]() Mon 5:09 - When I add Ozone 9 on Master, my CPU goes from about 50% to 110% and causes severe latency where sound is almost inaudible.

![]() Commercially available horse spleen ferritin (HSF), with its iron core removed (Millipore Sigma SKU# A3641), was chemically linked through solvent exposed native amino acids to a human Fab. Here, we describe a drug delivery system using ferritin to multimerize a Fab therapeutic. However, to the authors’ knowledge, there are no examples of adapting ferritin for delivery of multimerized antibody fragment (Fab) therapeutics via chemical conjugation. The latter can be achieved using molecular engineering techniques, including the addition of a peptide or protein tag (24). ![]() Thus, ferritin proteins are attractive vectors for the delivery of drug molecules, either through encapsulation in the central cavity, or through display of drug molecules on the external surface (23). In this case, the interaction was attained through ligand-metal chemistry (21,22), with the preparation of a 1:1 complex between cisplatin and the monomeric subunits of the human ferritin cage (11). More recently, it was used for the delivery of cisplatin, but without the use of an encapsulation protocol (20). Encapsulation of molecules such as cisplatin (12,13), carboplatin (14), oxaliplatin (15), ruthenium (16,17), and gold (18,19) has been achieved using various ferritin cage disassembly or reassembly procedures. Below pH 3, the ferritin assembly may then be dissociated and reconstituted (10), enabling drugs to be trapped inside the protein “cage” (11). While, in nature, the ferritin cavity is used to store and enucleate iron, it can also be repurposed for the encapsulation of drugs or dyes (9). Ferritin is made up of 24 subunits held together by non-covalent interactions, arranged in an icosahedral cage with 2, 3, and 4-fold axes of symmetry around a central cavity (7,8). Ferritin family proteins, found ubiquitously in nature, possess many attractive features for the delivery of biotherapeutics (3–6). There is a growing trend within the field of biotherapeutics to develop molecules with a high degree of valency (1,2). The combined results of this study confirmed that Ferritin–Fab conjugates were successfully generated. Following optimization of the conjugation strategy using LC–MS, in-depth characterization was performed using SEC-MALS-QELS. Here, we describe the design, production, and characterization of a multimeric ferritin–antibody fragment conjugate. The utility of molecular cages, such as ferritin and its derivatives for applications in drug delivery, is well-known. In mammals, ferritins are composed of 24 subunits that form an icosahedron with an external hydrodynamic radius of 6 nm, and an overall molar mass of approximately 474 kDa, depending on the biological tissue from which it is derived. Multivalent, self-assembling scaffolds such as ferritin, a ubiquitous protein found in most human cell types in addition to invertebrates, higher plants, fungi and bacteria, offer an attractive “natural” alternative to polymer-based scaffolds. Although this is traditionally accomplished using polymer-based molecular scaffolds, biocompatibility and the fate of polymer by-products are often of concern. The design of molecules with a high degree of valency can be useful for receptor clustering, T-cell recruiting, agonist activation, and half-life extension. ![]() It was the second time when Davies was offered the role at the age of 16 when his agent suggested the film and Davies later deeply personified his acting skills while battling with the transitions of the role like finding a girlfriend and accepting the world. Neither of those were terribly appealing options, so I went for door number three: download the macOS Mojave installer from the App Store. So, what’s a guy to do? I contacted Apple Support, whose less than helpful suggestions were either a) roll back to a Time Machine backup from before I enrolled in the Public Beta and then install the update (less than ideal, since I would lose any files created after that backup or modifications to other files), or b) do a full restore and start from there. I tried re-enrolling in the Public Beta program, restarting my Mac, then unenrolling and restarting again to see if it would point me towards the right update, but no dice. I’ve heard of others being stuck with dead-end builds of an OS and a tweet from Eric Holtam seemed to confirm that the build I was using, 18a389, wasn’t eligible for the update. I was fairly confident that the root of the issue here was something to do with having been in the Public Beta program. See, when I opened up the installer, I was met with another roadblock: an error message telling me that my Air “does not meet the requirements for this update,” with no further information. I’ve heard of issues like this in the past, so I cast about to find a download link to the standalone updater, which was graciously provided by Twitter follower Paul. ![]() Lo and behold, however, Software Update insisted that my current version of macOS Mojave 10.14 was the most recent, and no updates would be forthcoming, thank you very much. But there’s often some trickiness when it comes to getting off the beta–and this year, that’s where I hit a speed bump.
